In Vitro Preservation of Cymbidium ´ Pestrol Lady ´ PLBs through Moist Condition and Drying

Abstract
|
|
|
|
The experiment was conducted to investigate efficient methods in vitro for the preservation of protocorm like body (PLB) of Cymbidium ´Pestrol Lady´ through moist condition and drying (gradual dry, rapid dry). Excellent storage method of PLB was obtained from moist condition using the media with 3 g·L^(-1) hyponex and 4 g·L^(-1) peptone (H3Pa medium) in the dark. The optimal preservation temperature was 5℃ in drying storage and 10℃ in moist condition storage. The preservation periods according to the storage method were 6 weeks in gradual dry, 3 weeks in rapid dry storage and 18 weeks in moist storage condition without drying pretreatment. In drying storage method, fresh weight of PLB after storage and their regrowing plantlet survival after culture for 2 months were good when PLBs were dried for 30 minutes at 5℃. The condition of moist storage the medium was best storage methods for long term storage and also the drying storage method of PLB in a laminar flow chamber was useful methods for regrowth plantlet of PLB stored.
